S100A8 and S100A9 are pro-inflammatory members of the S100 family of secreted calcium binding proteins. They are produced by neutrophils and monocytes, and forms Ca2+-dependent heterodimer/heterotetramer complexes (known as calprotectin). Calprotectin (S100A8/A9) is upregulated at sites of inflammation (e.g. psoriasis, rheumatoid arthritis, cardiac ischemia). It is involved in facilitating leukocyte arachidonic acid trafficking and metabolism, modulation of the tubulin-dependent cytoskeleton during migration of phagocytes and activation of the neutrophilic NADPH-oxidase.
