Interleukin-18 (IL-18) is a secreted protein that belongs to the IL-1family. IL-18 is a proinflammatory cytokine and can be expressed by multiple types of cells, including endothelial and epithelial cells, keratinocytes, gamma delta T cells, and phagocytes. This cytokine can modulate both innate and adaptive. Once binding to IL18R1 and IL18RAP, it can form a signaling ternary complex which activates NF-kappa-B, triggering synthesis of inflammatory mediators. The combination of IL-18 and IL-12 can induce IFNG synthesis from T-helper 1 (Th1) cells and natural killer (NK) cells.
