Survivin is the smallest mammalian member of the IAP (Inhibitor-Of-Apoptosis) gene family. It regulates two essential cellular processes; inhibits apoptosis and promotes cell proliferation. Survivin also plays a main role in cancer progression because it prevents apoptosis by inhibition of caspase 3 and caspase 7, and by regulating the G2 and M phases of the cell cycle. Overexpression of survivin is associated with neoplasia and metastasis. Survivin might be used as a biomarker and a primary chemotherapeutic target for the detection and treatment of gastrointestinal tract cancer, especially esophageal, gastric, and colon cancers.

Post transnational modification: | Acetylation at Lys-129 by CBP results in its homodimerization, while deacetylation promotes the formation of monomers which heterodimerize with XPO1/CRM1 which facilitates its nuclear export. The acetylated form represses STAT3 transactivation. The dynamic equilibrium between its acetylation and deacetylation at Lys-129 determines its interaction with XPO1/CRM1, its subsequent subcellular localization, and its ability to inhibit STAT3 transactivation. |
Tissue Specificity: | Expressed only in fetal kidney and liver, and to lesser extent, lung and brain. Abundantly expressed in adenocarcinoma (lung, pancreas, colon, breast, and prostate) and in high-grade lymphomas. Also expressed in various renal cell carcinoma cell lines. |
